For final concentration and buffer exchange, and in some cases for the purification process as well, all Vivapure® kits require a benchtop centrifuge, pipettes and pipette tips.
In addition, Vivapure® Adenopack 500, Lentiselect 500 and Lentiselect 1000 require a peristaltic pump for viral vector purification.
For Vivapure® Adenopack 100, purification is performed using the supplied syringes. Alternatively, an optional tubing kit can be ordered separately for purification using a peristaltic pump.
Yes, Sartorius recommends and sells Watson-Marlow pumps for this purpose.
The Vivapure® kits take advantage of Sartobind® membrane chromatography for rapid anion exchange capture of viral vectors. The buffers supplied with the Vivapure® Adenopack and Lentiselect kits have been selected for optimal capture of adenovirus type 5 (Ad5) and VSV-G pseudotyped lentivirus, respectively. If your viral vector has an isoelectric point similar to one of these, the kits may be suitable. Otherwise, we recommend scouting and optimizing a purification process using the Sartobind® IEX Lab or Vivapure® IEX units, which are available to purchase separately.
