In this application note, we investigate the efficacy of recombinant growth factors in enhancing organoid expansion and allowing researchers meticulous control over culture conditions. Through the cultivation of mouse pancreatic and hepatic organoids in media enriched with Sartorius Research Use Only (RUO) Growth Factors and Cytokines, in combination with the Incucyte® Live-Cell Analysis System, we demonstrate their capacity to significantly enhance and sustain organoid growth for extended periods, outperforming other commercially available alternatives. Furthermore, these RUO reagents enable precise manipulation of concentrations, facilitating the identification of optimal culture conditions and the dissection of individual growth factor functions. This is exemplified through a concentration response curve applied to R-spondin1, showcasing their versatility in advancing organoid research.
This app note offers in-depth insights into the impact of growth factors and cytokines on organoid development, alongside real-time data analysis using the Incucyte® live-cell analysis platform. Together these tools allow understanding, precise control, and comprehensive optimization of organoid culture conditions and aid in the understanding of organoid growth dynamics.
