Viral-In-One: Complete Capture and Purification of Ad5 and VSV-G LV
Purifying viral vectors in research laboratories is critical for cell-based assays when assessing the suitability and efficacy of novel gene therapies.
Comprising Sartobind® membrane chromatography units, Vivaspin® centrifugal ultrafilters and all the required reagents, our range of complete kits support faster purification and concentration of adenovirus or lentivirus using minimal equipment, at high titres and with low endotoxin levels.
Fast and safe isolation of adenovirus type 5 (Ad5)
Fast and easy purification of VSV-G pseudotyped lentivirus
Viral vectors provide an efficient means of delivering genetic material into target cells and therefore play an important role in the development of new gene therapies and vaccines.
In gene therapy, these modified viruses allow precise targeting and manipulation of cellular functions, enabling the replacement of faulty genes associated with genetic disorders and diseases.
Viral vectors are also playing a growing role in vaccine development. For example, adenoviral vectors encoding the spike protein of SARS-CoV-2 were successfully used as a prophylactic vaccine against COVID-19.
Despite their promise in these applications, challenges remain, such as undesired immune responses against the viral vectors themselves. Using Vivapure® kits, researchers today are rapidly screening and developing the viral vectors that will be used in tomorrow’s therapies and vaccines.

All reagents, filters, membrane chromatography units and centrifugal ultrafilters needed to purify, concentrate and re-buffer your sample(s), depending on the kit ordered.
Vivapure® Adenopack can be ordered with or without Benzonase nuclease, depending on your requirements.

For Vivapure® Lentiselect, the kits and respective number of purifications are:
Yes, they are. While we have taken steps to minimize our environmental impact by removing printed manuals from the product packaging, the full instruction manual can be downloaded from the relevant product page in our eShop.
For final concentration and buffer exchange, and in some cases for the purification process as well, all Vivapure® kits require a benchtop centrifuge, pipettes and pipette tips.
In addition, Vivapure® Adenopack 500, Lentiselect 500 and Lentiselect 1000 require a peristaltic pump for viral vector purification.
For Vivapure® Adenopack 100, purification is performed using the supplied syringes. Alternatively, an optional tubing kit can be ordered separately for purification using a peristaltic pump.
Yes, Sartorius recommends and sells Watson-Marlow pumps for this purpose.
The Vivapure® kits take advantage of Sartobind® membrane chromatography for rapid anion exchange capture of viral vectors. The buffers supplied with the Vivapure® Adenopack and Lentiselect kits have been selected for optimal capture of adenovirus type 5 (Ad5) and VSV-G pseudotyped lentivirus, respectively. If your viral vector has an isoelectric point similar to one of these, the kits may be suitable. Otherwise, we recommend scouting and optimizing a purification process using the Sartobind® IEX Lab or Vivapure® IEX units, which are available to purchase separately.
Yes, you can. A comprehensive range of process chromatography and ultrafiltration solutions using the same membrane technologies supports an easy transition from R&D to commercial production.
