Downstream Vaccine Research
Vaccine research and development requires downstream process techniques to obtain the target antigen molecule and prepare the vaccine candidate for further analysis. The downstream process starts with the clarification and sterile filtration of small-scale cell cultures to obtain viruses and proteins in the supernatant, which is followed by concentration and purification of the molecules. This process can be very tedious and time-consuming.
To overcome this bottleneck, Sartorius offers rapid, user-friendly and scalable solutions to help you simplify cumbersome downstream application steps.
Sartorius Downstream Vaccine Research Solutions Offer:
Sartoclear Dynamics® Lab kits are designed for rapidly harvesting 15 mL to 1,000 mL volumes of cell culture in the lab, enabling clarification and sterile filtration in one step. Sartolab® ultistation enables hands-free and simultaneous filtration of up to six samples of small volumes.
Vivaspin® centrifugal concentrators provide near-total mAb, protein, virus and other nanoparticle recoveries, while achieving high concentration factors in the shortest time. Now, we offer both PES and RC membranes to optimize molecule-to-membrane compatibility.
- Unique PES and RC membrane range
- Fastest concentration times
- Angular dead stop for complete sample retrieval
- Market-leading technical and application support
Measuring vaccine potency or biologically active components is critical to the determination of a vaccine’s effective dose. In addition, the stability of the vaccine has a major impact on its usage for immunization programs worldwide. Octet® Bio-Layer Interferometry (BLI) systems deliver comprehensive characterization of vaccines as well as rapid and reproducible determination of protein concentrations for titer and functional activity assays. A single platform delivers high value across a wide range of applications in vaccine discovery, development and manufacturing.

Downstream Vaccine Research FAQs
Yes, Sartoclear Dynamics® Lab Kits with filter aid offer fast and effortless solution for clarification of sample volumes of up to 1L, enabling clarification and sterile filtration to be performed in one step.
It can! Sartorius provides a convenient conversion table to choose a MWCO up to 1/3 the diameter of the target virus. For example, high recoveries of Lentivirus - which has a diameter of ~90 nm - are achieved with 300 kDa MWCO Hydrosart® or 100 kDa MWCO PES membranes.
Ultrafiltration can be used for concentration, diafiltration, washing or purification of nanoparticles. In addition, ultrafiltration can be used to separate free- from nanoparticle-bound or encapsulated APIs. This enables a detailed understanding of nanoparticle functionalization, informing correct dosing while minimizing toxic effects.
Sartorius offers a comprehensive range of lab ultrafiltration solutions to meet the research and development needs for nanoparticles which will be used in tomorrow’s nanomedicines. Centrifugal, pressurized and crossflow devices enable processing of 0.1 mL to 5 L samples, offer superior process speeds compared to conventional techniques, and provide high target recoveries.
Use of centrifugal concentrators for buffer exchange or sample concentration requires optimization of the protocol and correct handling of the device in conjunction with a pipette to ensure:
- Determination of the concentrated sample volume for accurate yield measurement
- Retrieve the full volume of sample in a tall concentrator tube while avoiding sample-to-pipette and pipette-to-sample contamination
- Proper rinsing so that the preservatives used to maintain membrane stability do not elute into the sample leading to anomalous peaks in concentrate analysis

How does the quality of ultrapure water influence the data and reproducibility of virology research?
Many of the assays used in vaccine and virology research are vulnerable to interference and contamination. Ensuring the availability of RNase/DNase and endotoxin-free water is essential for accuracy. In addition, the purity of laboratory water can influence the quality of viral pseudotypes, with demineralized water significantly decreasing transduction capacity compared to ultrapure water.
Arium® water systems offer ultrapure water solutions suitable for sensitive analytical workflows such as those used in virology and vaccine research. Download the Arium Virus Research Application Note
